Gene synthesis is an efficient and cost-effective alternative to molecular cloning for custom gene production. AITbiotech provide comprehensive gene synthesis services including de novo gene synthesis, sub-cloning, mutagenesis and other customized request. Customers will receive ready-to-use plasmid for your downstream applications.
AITbiotech has established tight quality control steps during the whole duration of gene synthesis. Final plasmid will be sequenced (single stranded sequencing) to ensure 100% accurate sequence with desired. Free double-stranded sequencing service is available upon request.
Sequence is verified with Sanger DNA sequencing. The presence and size of insert is confirmed by restriction digestion.
AITbiotech is able to synthesize gene sequence from 50 bp to 20 kb, generally the primary
synthesized fragments range from 500 bp to 2 kb, but fragments could be further assembled to form large size.
Sequences with high GC content or repeats is complex to be handle, a higher cost and longer duration is required. We will carefully review every sequence, and provide a custom quote for every project, which includes turnaround time and price details.
We do not have strict criteria for “complex genes”. Based on our experience, a sequence will be considered as a “complex sequence” if it meets one or more of following criteria:
AITbiotech provide standard vectors such as pUC19, pUC57 and pBluescript II SK(+). Please take
note that synthesized gene is cloned into the pUC57 vector if not specified.
If not specified, synthesized fragments is cloned into pUC57 via Sma I or EcoR V, or any specific restriction sites within the Multiple Cloning Site upon request.
AITbiotech provide pUC19, pUC57 and pBluescript II SK(+) vectos. If you need to clone your
synthetic gene into a custom vector, please send us the vector DNA and pertinent information
(i.e. vector size, cloning site, copy number, etc).
AITbiotech will deliver 4.0ug plasmid DNA containing gene of interest in the vector.
The lyophilized plasmid maybe attached to the wall of the tube. AITbiotech will recommend
our customers to briefly spin down the tube before opening it.
The lyophilized plasmid is stable at -20°C for at least 1 year. Plasmid dissolved in TE is stable for at least 6 months at -20°C or 4°C. Plasmid dissolved in water is stable for at least 6 months at -20°C in the absence of nucleases. Please ensure the water used is at neutral pH to avoid depurination.
Inputting the effort, energy, and expertise, it is reasonable for the price of gene synthesis to be much higher than that of oligo synthesis. During gene synthesis, the oligos have to be assembled together in the correct order, which takes time and expertise.
Due to limitations in oligo synthesis chemistry and other practical considerations, oligos are produced as mixture of single-stranded DNA strands and are limited in length to ~200 bases.
Synthetic genes can extend to several thousand, double-stranded base pairs. Usually created as a sequence verified clone, the products from these constructs may be the best tool for your research needs.
